Amplification of tlh gene in other Vibrionaceae specie by specie-specific multiplex PCR of Vibrio parahaemolyticus Electron. J. Biotechnol.
Yáñez,Romina; Bastías,Roberto; Higuera,Gastón; Salgado,Oscar; Katharios,Pantelis; Romero,Jaime; Espejo,Romilio; García,Katherine.
Background The surveillance of Vibrio parahaemolyticus in the Chilean coast has been mainly performed by multiplex PCR amplification of three different hemolysin genes, which are specie-specific virulence factors. These genes are also employed in the determination of V. parahaemolyticus pathogenic load in seafood and for characterization of pathogenic strains associated to diarrhea cases in human. During environmental surveillance that we performed every summer, we occasionally observed a thermolabile hemolysin (tlh) PCR product of a slightly smaller size than expected, which was coincident with low loads of V. parahaemolyticus in the environment. In order to understand this observation, we probed the specificity of tlh primers for the detection of V....

Melo,Ligia Maria Rodrigues de; Almeida,Dulce; Hofer,Ernesto; Reis,Cristhiane Moura Falavina dos; Theophilo,Grace Nazareth Diogo; Santos,André Felipe das Mercês; Vieira,Regine Helena Silva dos Fernandes.
Ten out of fifty fresh and refrigerated samples of shrimp (Litopenaeus vannamei) collected from retailers in Natal (Rio Grande do Norte, Northeastern Brazil) tested positive for Vibrio parahaemolyticus. The Kanagawa test and multiplex PCR assays were used to detect TDH and TRH hemolysins and the tdh, trh and tlh genes, respectively. All strains were Kanagawa-negative and tlh-positive. Antibiotic susceptibility testing was done for seven antibiotics by the agar diffusion technique. Five strains (50%) presented multiple antibiotic resistance to ampicillin (90%) and amikacin (60%), while two strains (20%) displayed intermediate-level resistance to amikacin. All strains were sensitive to chloramphenicol. Intermediate-level susceptibility and/or resistance to...

Bactéries pathogènes indigènes des eaux estuariennes: cas des vibrios. Revue bibliographique ArchiMer
Fouche, Dominique.

Depuration Times of Donax trunculus and Tapes decussatus BJM
Çolakolu,Fatma Arik; Çardak,Mine; Çolakoglu,Serhat; Künili,Ibrahim Ender.
The present study was performed to determine the depuration time and ability of Donax trunculus (Wedge Clam) and Tapes decussatus (Carpet Shell) contaminated with Escherichia coli, Salmonella enterica subsp. enterica serovar Typhimurium and Vibrio parahaemolyticus. Clams were contaminated with each bacterium at the level of 7.0 - 8.0 Log10 cfu/g. After contamination, clams were analyzed every 3 h in the first 24 h time period and every 6 h until the 72nd hour. During the depuration process of both clams, the level of bacteria decreased quickly to 40% of initial load in the first 12 h. The results of this study indicate that the depuration time of carpet shells for all bacteria is 66 h. The depuration process of the wedge clam was different from the carpet...

We describe an outbreak of seafood-associated Vibrio parahaemolyticus in Galicia, Spain in on 18th of August 2012 affecting 100 of the 114 passengers travelling on a food banquet cruise boat. Epidemiological information from 65 people was available from follow-on interviews, of which 51 cases showed symptoms of illness. The food items identified through the questionnaires as the most probable source of the infections was shrimp. This product was unique in showing a statistically significant and the highest OR with a value of 7.59 (1.52–37.71). All the nine strains isolated from stool samples were identified as V. parahaemolyticus, seven were positive for both virulence markers tdh and trh, a single strain was positive for trh only and the remaining strain...

MRNA detection by reverse transcription-PCR for monitoring viability and potential virulence in a pathogenic strain of Vibrio parahaemolyticus in viable but nonculturable state ArchiMer
Coutard, Francois; Pommepuy, Monique; Loaec, Solen; Hervio Heath, Dominique.
Aims: This work investigates the maintenance of viability and potential virulence of Vibrio parahaemolyticus in a viable but nonculturable population (VBNC) state by reverse transcription-polymerase chain reaction (RT-PCR). Methods and Results: Housekeeping genes, 16S-23S rDNA and rpoS, as well as virulence genes, tdh1 and tdh2, were selected and detected by PCR in a pathogenic strain of V. parahaemolyticus (Vp4). Their expression was then studied by RT-PCR in V. parahaemolyticus Vp4 cultivated in rich medium at 37degreesC. The 16S-23S rDNA and rpoS, tdh1, tdh2 genes were transcripted at the mid-logarithmic, stationary and late stationary phases, corresponding to various physiological states. The expression of these genes was also studied by RT-PCR in a...

Recovery in culture of viable but nonculturable Vibrio parahaemolyticus: regrowth or resuscitation? ArchiMer
Coutard, Francois; Crassous, Philippe; Droguet, Michael; Gobin, Eric; Colwell, Rita; Pommepuy, Monique; Hervio Heath, Dominique.
The objective of this study was to explore the recovery of culturability of viable but nonculturable ( VBNC) Vibrio parahaemolyticus after temperature upshift and to determine whether regrowth or resuscitation occurred. A clinical strain of V. parahaemolyticus Vp5 was rendered VBNC by exposure to artificial seawater ( ASW) at 4 degrees C. Aliquots of the ASW suspension of cells ( 0.1, 1 and 10 ml) were subjected to increased temperatures of 20 degrees C and 37 degrees C. Culturability of the cells in the aliquots was monitored for colony formation on a rich medium and changes in morphology were measured by scanning ( SEM) and transmission ( TEM) electron microscopy. Samples of VBNC cells were fixed and examined by SEM, revealing a heterogeneous population...

The use of API 20E for the identification of Vibrio anguillarum and V. ordalii OMA
Grisez, L.; Ceusters, R.; Ollevier, F.P..
Sixty-eight strains of Vibrio anguillarum, five of V. ordalii and the type strains of V. alginolyticus, V. carchariae, V. damsela and V. parahaemolyticus were compared using the API 20E gallery. Within the V. anguillarum strains, distinct groups could be separated mainly on the basis of their reaction on indole production and the fermentation of amygdalin and arabinose. Vibrio ordalii, the former V. anguillarum biotope 2, could easily be separated from V. anguillarum and from the other fish pathogenic Vibrio spp.
